If the data on the PC is critical to your business then the first thing you should do is attempt to back up the important data some how.

• You said you cannot use your USB port, but can you burn the data you need to a CD/DVD.
• How about taking out the harddisk and putting it into another well protected PC as a slave drive and copying off the files you need?
• You could also run scans on the problem hard disk while in the other PC to see if anything related to the malware can be found and removed.

Other possible options (but backup of important data should be completed first if possible):

• Try using System Restore to restore to a restore point before the problem began to see if you have at least more functionality so that you can run our cleaning procedures ( READ & RUN ME FIRST. Malware Removal Guide )
• Use another PC to make a special CD which you can boot from to try and run virus and spyware scans or to at least backup data. CDs like the below:
