My iTunes library won't play following iTunes download

I bought and downloaded an album from iTunes store. The download was fine and plays OK but since then my existing library, which is made up of my CD collection and purchased items from iTunes, won't play and I am directed to "locate" my music from elsewhere in my computer. If I do this I can then make that track play but as I have some 4500 tunes in my library this will be very time consuming. Have I inadvertently turned something off? Also when I look at the "info" for the track no route is shown and it says it's "not available".
Any ideas would be much appreciated.

As far as I am aware, you must have somehow, maybe not on purpose, moved those music files to anoher location on your hdd. I am unaware of any way bar selecting the file root manually for all 4500 tracks, maybe someone else have the answer for you!
