Virus problem...

Im not very good with computers and all so bare with me. Basically i have some type of virus or something on my computer that doenst allow me to acces and file or anything, whenever i click on an icon or a file it just says "Windows cannot access the specified device, path, or file. You may not have appropriate permissions to access the item." I have tried installing a few virus removal programs, and they will be able to be accesed as long as i do not close the window or shut down my computer, once i shut down my computer that same error message comes across. any help or advice would be greatly appreciated
