Ran Disk Defragmenter. Completed successfully, but message at end said "some files could not be defragmented." Is this something I need to address?

theres reserved space for certain system files. that reserved space dosnt get defraged, so that might be what its talking about.

There are files that can't be defragmented.
Frankly, I don't see any sufficient reason for defragmentation nowadays. Now the drive sector size is too small compared to the size of files. Besides, now repositioning of HDD head is fast enough, which makes defragmentation pluses very doubtful.

Download (google for) pagedefrag from SysInternals (now M$)
to defrag your page file, which isn't usually accessible by the
    regular windows tool.

Some files, like movies which are being downloaded may be in use, and thus can't be moved.

what about a large file that is used in a game i play. as far as i know, it isn't being used at all. but whenever i go to defrag my hard drive, it says "some files cant be defragmented" and it lists that file. The file itself is about 6GB and its basically the entire game.
Please explain what i can do to fix this. the game is World of Warcraft.

But, if there isn't a contiguous 6 GB's (or more) of free space to move that file, that file will not be defragged...

You can defrag WoW files using PerfectDisk 8.0. If it is your system drive, you can have it do an offline defrag, where it optimizes free save. Then most of your Wow files may have been defragged. The last file that is not defragged, can be defragged using the "single file defrag" utility in Perfect disk. It will move out files to specifically make room for common.mpq
