help with first multi hd setup please

hi everyone i just put together my shinny new game / work rig with an ssd 90 gb to run my OS and a 250 seagate drive for storage and to run most applications from. but how do i setup my larger HD to run games and applications the seagate drive is from my previous machine. everything i try to run off it right down to yahoo messenger says "side by side configuration is incorrect! ..i'm sorry for the noob cuddle guys if i knew what to search for i'd be doing that instead.
