For the best security, I would start with the BIOS setup, making a BIOS password. That will keep it mostly safe while you are away with it off. They would have to know how to circumvent it or guess the password.
When you leave while it is on, you can set a password for the screensaver. I have put a shortcut to a screensaver on the desktop to run while I was away before, so I could make sure it was running when I was gone. There are other things you can do, also, but this is a good start, and will make it hard for most people to get access. As long as there is always a password guard active while you are gone, they will have to deal with it, at least stalling the attempt.
One other thing you can do is make a user with minimum privileges and use it generally, in case you forget to put on the screensaver or something. Then only use the main one when you can be there the whole time. It is still possible for them to use the minimum privileges to do stuff, but not as bad as starting in the main account.
