You need to have a password protected user logon, and a screensaver that comes on in 5 minutes. Without logging in, no one can access your desktop. If you are really this concerned, then the inconvenience will not matter. If you leave your station for a break, log off.

If you're on a networked system in a larger office, address your concerns to IT. I don't see how people can snoop your computer if they don't walk up to it when you aren't there. If you leave it logged on and running all the time, you're just asking for the problem if you can't trust your co-workers.
