Would making a copy of the system32 directory as well as saving a copy of the registry make for a good enough system restore?

So if say you got yourself utterly infected with viruses up to your nose, you just drop in the backup ups. Not that it would get rid of all the infected software, but it would kill any ties to the system, so they wouldn't activate again right?

Agreed with Coleman, backup in a conventional way as doing that may lead to more system issues than just being malware infected.. old file version, new added ones, re-registering is a possible of many dlls, etc plus depending on Windows version, that location and many folders are system locked and taking ownership would be a pain as well as a potential security risk.

Many freeware backup utilities are available here http://majorgeeks.com/downloads3.html (other licence software will be amongst so look at licence) but noteables are DriveImage XML and Karen's Replicator
