Memory loss...?

My friends computer is a ASUS CUSL2 MB.I've messing with it for the last few days but this question is more for any MB with more than 2 memory slots. His Mb max's out at 512 for memory but has three slots. Is it better to use ALL thre or is it ok to use 2 256's in 2 slots? I been meaning to ask this forever but keep forgeting.

2 x 256 will work just fine. As should 1x512 for that matter.
