How can I get the number keys on the far right of the keyboard to work?

There is a key (probably in the top left of the number pad) with Num Lock written on it. This key toggles whether the keys are used as arrows for cursor navigation. There may also be a light on your keyboard that indicates the Num Lock Status.

As Matt posted, press the NumLock key.

But it is also part of the boot/bios to set it on at start up.

The bios setting you are refering to determines the status of the numlock key at startup, either Numlock ON or Numlock Off. The setting won't make much difference to its use in windows as you will only have to toggle it.
