Lots more info needed: brand model of usb wifi adapter, brand and model of router, any security on the router, what OS is the computer using the wifi adapter.

Did you release IPs between using it in the laptop and then plugging it in your desktop? The router has leased the wireless to the laptop and the desktop is a different computer.

    ipconfig /release
    followed by ipconfig /renew

Are you using Windows or the manufacturer's software to configure the connection? Sometimes I find that BOTH need to be utilized to connect successfully. Is the driver properly installed?
